Deploying the model of knee osteoarthritis in wistar rats

Abstract
Objective: Knee osteoarthritis model was induced in rats with synovial injection monosodium iodoacetate (MIA).
Subjects and methods: 9-week-old wistar rats were injected with 1.0mg/30µL MIA into kneejoint to cause knee osteoarthritis. The control group was injected with physiological saline. Evaluate the change in the weight bearing with a specialized measuring device (Incapacitance tester); serum PGE2 and proinflammatory cytokines TNF-α, IL-1β, and IL-6; and histopathology of the knee.
Results: Compared with the control group, rats in the MIA injection group had a reduction in the weight bearing (p < 0.001); serum concentrations of PGE2, TNF-α, IL-1β, and serum IL-6 were increased (p < 0.001); knee-joint histopathology showed clear osteoarthritis with manifestations: the knee joint has thick synovial membrane, infected with many inflammatory cells including lymphocytes and macrophages; in some places the synovial membrane has fibrotic necrosis; congestive blood capillaries; cartilage tissue is degenerative and hyalinic.
Conclusion: The model of knee osteoarthritis has been successfully deployed in wistar rats by injecting MIA (monosodium iodoacetate) 1.0mg / 30µL.
