Quanlitave, quantitative methods for Ophiopogon japonicus (L.F.) Ker-gawl herbal grown in Vietnam

Abstract

Objective: To establish a qualitative thin-layer chromatography (TLC) method and a quantitative UV–Vis spectrophotometric method for Ophiopogon japonicus.


Subjects and methods: Ten samples of Ophiopogon japonicus tuber roots were collected from different regions. TLC and HPLC–ELSD methods were applied to qualitatively determine the presence of Ophiopogonin D and Ruscogenin, while the UV–Vis spectrophotometric method was used to quantify the total saponin content, expressed as Ruscogenin equivalents.


Results: Several samples showed no detectable Ophiopogonin D spots on TLC or HPLC–ELSD chromatograms. However, after acid hydrolysis, all samples exhibited peaks corresponding to the Ruscogenin reference standard. Quantification by UV–Vis revealed total saponin contents ranging from 0.51% to 2.19% (as Ruscogenin equivalents). Conclutions: The TLC method comparing acid-hydrolyzed samples with Ruscogenin, together with the UV–Vis spectrophotometric quantification of total saponins expressed as Ruscogenin equivalents, provides a simple, cost-effective, and rapid approach suitable for quality evaluation  of Ophiopogon japonicus raw materials.
