Acute toxicity and analgesic effects of Khop Bao An capsules in experimental animals

Abstract
Objective: To evaluate the acute oral toxicity and analgesic effects of Khop Bao An (KBA) capsules in mice.
Subjects and methods: KBA’s acute toxicity and analgesic effects were assessed according to Ministry of Health guidelines and using hot plate and acetic acid-induced writhing tests.
Results: At the highest dose that mice could ingest, 26.42g of mixed dry extract per kg, KBA did not cause deaths. All mice remained healthy and showed significant weight gain on day 7 compared to that on day 0 as well as had normal visceral organs under macroscopic examination. In addition, at doses of 989.28 and 2,967.84 mg of mixed dry extract/kg/day for 7 consecutive days, mice were able to notably prolong the duration of pain tolerance to heat stimulation compared to those before treatment and the control group. Also, the numbers of mice’s writhing episodes at each 5-minute interval within 30 minutes after injecting acetic acid were remarkably reduced, and the times to the first writhing episode were longer than those of the control group.
Conclusion: KBA did not cause acute toxicity in mice and had both significant central and peripheral analgesic effects at the tested doses.
